People have gotten hot on a neurotoxic metabolite again, citing the fact (which I don't think is a fact), that direct application of MDMA to the brain is not neurotoxic.
Do you think mdma is neurotoxic? If so, what do you think causes it?
If that is the case then how does this system work? Also, once the molecule of mdma enters the neuron via uptake transporter, does it exit back through the transporter. Is the serotonin release only from the reverse uptake or do the viscules also merge with the cell wall and release. If mdma is causing the reversal of the transporter to release serotonin into the synapse, how does the serotonin leave from the viscule into the neuron. In other words, what kicks it out of the viscule so that its just floating around in the neuron? the mdma?

MDMA is definatly neurotoxic at the doses they give to experimental animals... I don't know about humans.
What causes it? Probably a mix of dopamine degredation by MAO-B, probably MDMA itself somehow releases free radicles... *shrug* I don't know.

I don't quite get your next paragraphy.

MDMA may exit again through the transporter, it probably accumlates in the vesicles largely. Vesicles keep merging just like normal. There is always serotonin floating around, thats where it gets put into vesicles from.

I did this animation, if you've got powerpoint. I got kinda lazy at the end, but I think you can figure it out.

It's surprisingly complicated eh? It's really weird, the majority of that whole thing was figured out in two papers...
 
Just saw the animation. I didn't understand any of the first bit. Maybe in time. I see what your saying about the viscules, under the influence there is just tons of serotonin everywhere. Probably why my question above didn't make any sense. Is there a large increase in the rate in which the viscules merge with the cell wall while under the influence of mdma as compared to normal conditions?

I spend a large portion of my day riding around in a delivery truck (as a passenger) so I have plenty of time to read up on this. What sort of things could I read to help me develop a better understanding of brain chemistry. A good starter read?

Well it depends on whoes theory you look at. I suspect largely there is no increase in the rate of vesicle docking, or a decrease because of autoreceptor action.

Some people like Molecular Neuropharmacology. I especailly don't like The Biochemical basis of Neuropharmacology. I've never really dug any undergrad neuropharmacology text. One of the best texts (but also possibley a bit advanced), is available online here.
