What factors affect clinical outcome of elevated plasma levels??

Talking specifically combination of enzyme inhibitors, with enzyme substrates.

Cmax can triple or even quadruple, area under the curse increases exponentially - but it's noted that no dose reductions are necessary.

I get the answer - "there's no simple answer" - but, just to garner some degree of insight?

How could that possibly be the case?

Specifically I'm talking Atomoxetine with an SSRI like paroxetine or fluoxetine.

Plasma levels go way way up, but supposedly don't impact clinical outcome.

So - it's obvious not directly like taking 3 or 4 times the dose of the inhibited drug so - what then?

Is there a way to find some kind of an answer to this question?
 
Do you have a source for the claim that "no dose reductions are necessary"? Because that seems like a pretty fucking broad statement.

At any rate, the thing about many psych meds is that there is great interpersonal variability in regards to dosage. One person might do well on 75mg of venlafaxine, another patient may need 300mg (or more). Also, dose-response-curves tend to be non-linear.
