As you can see the structures are very closely related.
Amantadine appears to have a very wide receptor activity profile. Hitting NMDA sites, increasing dopamine release and blocking its reuptake, and affecting some anticholinergic sites. Amantadine has a significantly shorter half-life than Memantine. Amantadine is fully excreted after 15 hours for a 100mg dose or a rate of 5 mg per hour. Maximum blood levels of Amantadine are reached within four hours for a 100mg dose. (This information is from the monograph for Amantadine found
