I'm wondering about the interaction between lymecycline (a member of tetracycline antibiotics) and methadone, and clonazepam. I mean how the drug is metabolized or rather I'm interested in induction of any enzymes because it's quite clear how it is metabolized from looking at the structure.

The leaflet says nothing about interactions with opioids and benzodiazepines, it actually says very little about interactions. I've searched the net, tried seeking some info in patents, on some forums, well, generally whatever would come up. I know there's a thread here about MDMA and antibiotics interactions but it doesn't list lymecycline and it actually differs from a simple tetracycline having a chain on the amide group ending with a carboxyl group. It changes a lot compared to tetracycline (solubility at any pH, >95% BA after oral administration), so it may be also different in acting on enzymes. It's definitely a substrate of CYP3A4 and this subtype is very important in metabolism of both methadone (to normethadone) and clonazepam (to 7-aminoclonazepam among others).

The problem is I already have a problem with dosing methadone - I have to do it twice daily not to feel any withdrawal symptoms. I can't tell by subjective feelings because I've just started the treatment with lymecycline (3 300mg doses and I'm already feeling weak:|) as my nightmare from teenage came back (I suspect it has to do with skin exposure to some chemicals and I've always had problems with skin during my adolescence, probably a genetic thing but whatever). Well, if it was only a substrate of CYP3A4, then it wouldn't do me any harm but if it's an inducer then it'll be a problem because I'm tapering down methadone.

So, does anyone have some access to some articles that might clear this up or maybe have any info from other sources?
Thanks in advance.

It seems like it does have "bad" impact on methadone metabolism. I took a dose that usually made me a bit high before I started the treatment with lymecycline and I felt like I took a regular dose. And again I started to feel pain in my gut and a "prelude" to diarrhea (the beginning of methadone withdrawal starts with a pain like something twisted your intestines and stool is hard...).

I don't like it because I've got the script for a higher dose and I take 2 doses a day which is a dose for rosacea and not acne vulgaris + another antibiotic in gel and adapalene in ointment. I thought 4 days would result in some improvement like no new spots. And I'm just weak from taking the antibiotic and I'm scared of stopping it now as the shit will probably hit back even harder if I stop taking the antibiotic.

Meanwhile I found some article stating that tetracycline is either just a substrate of CYP3A4 or a substrate and an inhibitor. I don't feel it with lymecycline. There was a day I totally skipped my dose of clonazepam but that proves nothing because it was a day I spent almost entirely in bed and besides I took more clonazepam than my dose is now the day before and even if I didn't, this is easy to do when you've been for years on a benzodiazepine with such a long half-life...
