I spoke to someone who has experience smoking or vaping 5-MeO-DMT and who also tried it orally in combo with B. caapi extract. With the oral experiences, he experienced "definitely a euphoric mdma type effect" and no ego loss. I asked him if he normally gets that effect from 5-MeO and he said, "maybe a little but 5meo vaped hits so fast it’s kind of hard to tell before you go to that impossible to explain...". I haven't done 5-MeO-DMT, but I have done 5-OH-DMT, more commonly known as bufotenine. Although all of my snorted doses were too low to be formidable, when I consumed a few droppers of bufotenine tea after ingesting harmine, I was pleasantly surprised to feel what felt like MDMA and I suspect that this is not a typical effect.

5-MeO-DMT is a serotonin reuptake inhibitor and 5-OH-DMT is a serotonin releaser and 5-MeO-DMT partially metabolizes to 5-OH-DMT. This indicates that these chemicals synergize with MAOIs. I also consumed ¼ of a Zoloft after consuming harmine and got an undeniable, brief, MDMA-like effect, certainly distinct from the effects of either drug on its own. This confirms the synergy between the two mechanisms.

In mental depression, new approaches could also combine both MAO inhibition and serotonin reuptake inhibition to increase extracellular 5-HT concentration at the synapses.

This is an incredibly intriguing prospect as 5-MeO-DMT and harmine (perhaps also 5-OH-) have been shown to induce neurogenesis, unlike MDMA; and they're legal.

I made another post where I provided more detail, including references for the above quotes and claims: Herbal ecstasy is real
