Does nephrotoxicity depend on quantity of a drug?

I have this idea that for instance if you have to take 5g phenibut or 0.5g pregabaline, the kidneys probably have to work a lot harder to excrete the 5g than the 0.5g.

Is this true? Does nephrotoxicity depend on quantity? Are all substances that get excreted by the kindneys toxic at a certain concentration? Or is the shape and function of a molecule more important then the quantity of the molecule?


Finnaly: is nephrotoxicity a known side effect of phenibut?
