There seems to be no definitive answer really anywhere on the internet or in circles or peer reviewed journals.

Have any long-term studies been done or are most current information purely conjecture?

If someone were to use proper oxygen, vitamins, and nutrients in diets, how much or how frequently can nitrous oxide be used without causing the infamous nerve damage, hypoxia, shakes, or mania?

Plenty of research has been done on the methods by which the damage occurs, but it's not really feasible to answer that question specifically. The range is going to vary too widely from person to person, and most studies are done using a continuous supply of N2O rather than the more typical random pattern of inhalation followed by recreational users. In order to mimic the typical usage pattern, you'd have to do too many test cases. It's certainly not impossible to figure out a reasonable range for most people, but there isn't a great scientific reason to do it, hence the lack of info.
