1 time use of methadone now what?

A friend has been taking 30-40 mg tabs or percs a couple times a day to deal with severe back pain. They got a suboxone film last week to try and was warned to not take it all at once because it might make them sick. They took 1/4, then another 1/4, and finally after taking the rest they felt relief and maybe a little bit of a high. Not able to find any tabs this week, someone mentioned methadone to them and they bought a 40mg waffer to try. Took half of it yesterday around 5pm and never felt the first bit of relief or any other kind of effects. Now they have a few tabs and a couple subutex pills (they have never tried these) but are unsure what their next move should safely be after doing their research on methadone and how it stays in your system for so long. Since this is their only time using methadone and never even felt an effect from it should they still wait a couple days before taking anything else?
